Bis zu seiner Pensionierung im Jahr 2024 war John Comaroff Hugh K. Foster Professor für afrikanische und afroamerikanische Studien und für Anthropologie an der Harvard University. Bis 2012 war er der Harold H. Swift Distinguished Professor of Anthropology an der University of Chicago.
Ausgebildet an der University of Cape Town (B.A., 1966) und der London School of Economics (Ph.D. 1973), forscht er in Südafrika zu Kriminalität, Polizeiarbeit, Persönlichkeit und der Funktionsweise des Staates, Demokratie und Differenz sowie vorkolonialer, kolonialer und postkolonialer politischer Ökonomie und Recht. Er hat auch über ethnische Zugehörigkeit und Identität, afrikanische Wirtschaft, Gesellschaft, Religion und Kultur geschrieben.
